Naturally immortalized human breast carcinoma KAIMRC1 cells with Immuno-phenotypic characteristics that grow in 3-D

Objective

A continuous human breast carcinoma cell line (KAIMRC1) has been established and characterized. These cells are epithelial like and cuboidal in shape. The epithelial nature of these cells was confirmed by flow cytometry analysis. Interestingly, in normal cell culture conditions these cells show capability of growing both in monolayer as well as in 3-D conformation. Phenotyping revealed stemness of these cells. Immunocytochemistry and western blot analysis showed that these cell are ER, PR positive and HER2 negative. They exhibited a modal karyotype of 58-63, X with abnormalities in chromosomes 3 and 9 and an absent chromosome 17. Several assays were conducted to monitor the growth behavior, wound healing, colony formation in soft agar and mammosphere formation of the cell line. KAIMRC1 cells found to be more responsive to drug treatment in comparison to established MDA-MB231 and MCF-7 cell lines. Whole genome and exome sequencing are underway to dig deep into the genetic and environmental factors involved in the development of this cell line.Moreover, KAIMRC1 grow in the 3-D form in normal growing media and better than any other cell lines tested with unique immune-profile that could lead to the discovery of novel drug targets.In conclusion we have isolated and characterized a new naturally immortalized cell line, KAIMRC1 and now characterizing another naturally immortalized cell line, KAIMRC2. We envisage that these cell lines will play a key role in opening up new doors toward the understanding of breast carcinoma.
