Novel T cell-based tools in diagnosis and management of microbial infections

T cell diagnostics is an emerging field addressing essential clinical
needs relevant to transplantation medicine, infectiology and autoimmunity. Lophius
has developed innovative technology platforms (T-activation® and Reverse T cell
Technology - RTT) permitting a sensitive quantification of functional
pathogen-reactive effector and memory T cells. The T-activation® technology,
based on a proprietary antigen formulation, was transferred into the development
of a highly sensitive, standardized, reliable and user-friendly IFN-γ ELISpot-based
immune monitoring tool, leading to the development of a self-declared CE-marked
IVD kit (T-Track® CMV) according to IVD directive 98/79/EC. T-Track® CMV allows
the sensitive and
reliable monitoring of functional impairment of T cell-mediated immunity in
immunocompromized patients at risk of CMV reactivation and CMV-related clinical
complications. Two completed and one on-going prospective multicenter clinical
validation studies in solid organ and allogenic hematopoietic stem cell
transplant recipients established correlates between T-Track® CMV results and clinical outcome, thus
demonstrating the principal suitability of T-Track® CMV in
assisting clinicians to evaluate the risk of CMV disease and to guide therapeutic choices. An
ongoing interventional study in a cohort of recipients of solid organ
transplantation with high risk of CMV-related complications aims to assess
health-economic benefit in addition to clinical outcome.
