Stability of DMSO-solubilised compounds: The effects of temperature and humidity

As well as identifying how compounds degrade, the rate at which these transformations occur upon storage may have considerable impact on all supported assays in early pharmaceutical development. Several studies were conducted to investigate the stability of compounds solubilised in DMSO and to characterise the rate of degradation with respect to temperature and humidity. Most compounds degraded significantly within the first 6 weeks of solubilisation and compound purity was best retained under frozen conditions stored at -19 °C where degradation was considerably suppressed. Contrary to our initial predictions, the influence of controlled atmospheric conditions at 10% relative humidity was found to exert no significant effect on compound integrity. Where large investment in controlled humidity is a strategy undertaken by many Pharmaceutical and academic institutions, these data may shape future decisions for optimising small molecule storage conditions to best retain compound purity in a cost effective manner.
